产碱性纤维素酶海洋细菌的筛选、鉴定及酶学性质研究 被引量:22

摘要 用CMC平板筛选方法,从青岛近海海域海水中分离出一株产碱性纤维素酶的海洋菌株QM11,经16S rDNA鉴定,该菌株为Cytophaga fucicola。对该菌的生物学特性研究表明,其最适生长温度为27℃,生长温度范围为4-48℃,为耐冷菌;在pH7.0~8.0、含3.0%NaCl的培养基条件下,最适宜菌株生长和产酶;QM11所产碱性纤维素酶最适反应温度为40℃,最适反应pH为9.0,在碱性条件下具有较高的酶活性和较好的稳定性。Mn^2+、Fe^3+对酶反应具有促进作用,Cu^2+、Pb^2+对酶反应具有抑制作用。 Using CMC plate screening method, a marine strain QMll that can produce alkaline cellulase is isolated from the seawater of Qingdao coast. According to the 16S rDNA analysis, the strain belongs to the Cytophagafucicola. Studies of its biological characteristics have shown that the optimal growth temperature is 27 ℃,and the growth temperature range is between 4℃ and 48 ℃ ,which indicates it is a cold-resistant bacterium.Media of pH7.0-8.0 and 3.0% NaCI are the most suitable for the growth and enzyme production.The optimal temperature for QM11 producing alkaline cellulase is 40℃.The best reacting pH is 9.0,and the strain has a high enzyme activity and a good stability in the basic conditions. Studies also show that Mn^2+ and Fe^3+ could stimulate the enzyme reaction, while Cu^2+ and Pb^2+ inhibit the reaction.
