摘要
目的:观察水蛭和虻虫不同剂量配伍对血瘀大鼠血液流变学的影响,探讨药对剂量配比的合理性。方法:将大鼠随机分为空白对照组、模型对照组和4个不同剂量比的水蛭、虻虫配伍组(水蛭∶虻虫分别为3∶4,4∶3,3∶1,6∶1,总剂量均为1 200 mg.kg-1)。连续给药7 d。于d 6给药1 h后,除空白对照组外,其余各组以皮下注射盐酸肾上腺素及冰水浴复制大鼠血瘀证模型。次晨给药后2 h采血,测定血液流变学指标。结果:不同配比的水蛭、虻虫药对均能明显改善大鼠血瘀模型的血液流变学异常,其中配伍2组(4∶3)、配伍3组(3∶1)对血细胞比容、红细胞聚集指数的作用较强。配伍3组(3∶1)对血浆黏度、卡松黏度的作用较强。配伍3组(3∶1)、配伍4组(6∶1)对血全黏度和全血还原黏度作用较强;配伍4组(6∶1)对红细胞变形指数作用较强。结论:在总剂量相同的条件下,水蛭剂量相对较大的配伍组(3∶1,6∶1)对血液流变学指标的作用较明显。
Objective :To observe hemorheological effect of the different combinations of Hirudo and Taba- nus on rat models of blood stasis syndrome, and explore the rationality of different ratio of Hirudo to Tabanus. Methods:Rats were divided randomly into 6 groups: blank control group, model control group, Hirudo and Tabanus combination groups ( ratios of Hirudo to Tabanus were 3: 4,4:3,3 : 1,6: 1, respectively, and their total dosage was 1 200mg·kg^-1 ). The Hirudo and Tabanus were administrated by gastric gavage, qd, for 7 consecutive days. Rat models of blood stasis syndrome were established by subcutaneous injection of adrenaline combined with the stimulation of ice water. Results : Different ratios of Hirudo to Tabanus markedly improved hemorheological parameters on rat models of blood stasis syndrome. The group with the combination proportion of 4:3 and 3:1 showed a better effect on hematocrit and erythrocyte aggregation index. The group with the combination proportion of 3:1 showed a better effect on plasma viscosity and casson viscosity. The group with the combination proportion of 3:1 and 6: I showed a better effect on blood viscosity, blood reduced viscosity. The group with the combination proportion 6:1 showed better effect on erythrocyte deformation index. Conclusion:In the different combinations groups, the group with the combination proportion of 6:1 and 3:1 showed better effect.
