雷帕霉素对柯萨奇病毒B3诱导的大鼠心肌细胞Ⅰ型胶原表达的调控

Effects of rapamycin on collagen type Ⅰ expression in rat myocardial cell induced by CVB 3

摘要 目的:研究雷帕霉素对柯萨奇病毒B3(coxsackievirus B3,CVB 3)诱导的大鼠心肌细胞Ⅰ型胶原表达的影响,探讨雷帕霉素治疗病毒性心肌炎的可能机制。方法:用CVB 3感染原代培养的SD大鼠心肌细胞,取浓度为10 nmol.L-1的雷帕霉素进行干预,采用RT-PCR和W estern B lot检测细胞Smad 3及Ⅰ型胶原表达水平。结果:正常心肌细胞未见Ⅰ型胶原表达,CVB 3诱导心肌细胞Smad 3和Ⅰ型胶原表达,雷帕霉素抑制其表达(P<0.05)。结论:心肌细胞在病毒性心肌炎(VM)时参与胶原调控,雷帕霉素通过Smad信号途径抑制Ⅰ型胶原表达。 Objective:To observe the influence of rapamycin on collagen type Ⅰ expression in rat myocardial cells induced by eoxsackievirus B3 ( CVB 3 ) and investigate the possible mechanism of treating viral myocarditis by rapamycin. Methods: Primary cultured SD rat myocardial cells infected by CVB 3 were treated with rapamycin in 10 nmol · L^-1. The expressions of Smad 3 and type Ⅰ collagen were detected by RT-PCR and Western Blotting respectively. Results:There was no expressions of collagen type Ⅰ in normal myocardial cells. CVB 3 up-regulated the Smad 3 and collagen type Ⅰ expression in myocardial cells, however rapamycin inhibited their expressions (P 〈 0.05). Conclusion : Myocardial cells may participate in the regulation of collagen in viral myocarditis and rapamycin inhibits the collagen expression through the Smad signal pathway.
