摘要
目的探讨肿瘤抑制基因RASSF1A在骨肉瘤组织中的表达与骨肉瘤的生物学行为之间的关系。方法用RT-RCR方法检测a)骨肉瘤MG-63细胞中RASSF1A基因的表达;b)30例骨肉瘤组织、10例骨软骨瘤组织及10例正常骨组织中RASSF1A基因的表达。结果a)骨肉瘤MG-63细胞中未发现RASSF1A基因表达;b)30例骨肉瘤组织中RASSF1A基因缺失率为43.33%,对照组骨软骨瘤中表达率为100%,正常骨组织中表达率为100%。RASSF1A基因的表达与肿瘤的Enneking分期及是否发生远处转移相关(P<0.05),而与患者的年龄、性别、肿瘤的部位及大小、Price组织学分级及是否侵犯软组织无关(P>0.05)。结论RASSF1A基因可能在骨肉瘤的发生发展中起重要作用,有可能为骨肉瘤的基因治疗提供新的靶点。
Objective To investigate the expression of turmor-supressing gene RASSF1A in osteosarcoma and its correlation with biological behavior. Methods To detect the expression of RASSFIA gene in osteosarcoma cell line MG-63, 30 cases osteosarcoma issues, 10 cases osteochondroma issues and 10 cases normal bone issues with the method of RT-PCR. Results (1)There was no expression of RASSFIA gene in osteosarcoma cell line MG-63. (2)The expression negative rate of RASSF1A was 43.33% ;no expression absence of RASSF1A gene was detected in osteochondroma and normal bone issues as control groups. The RASSF1A expression was significantly associated with the Enneking's stages and distance metastasis (P〈0.05), but not with age, sex, tumor locus, size, Prices' s grade and soft tissue invasion(P〉0.05). Conclusion Absent expression of RASSF1A gene in osteosarcoma may play an important role in tumorgenesis and tumor progression,and it may provide new target for gene therapy.
