δ-连环素在糖尿病大鼠海马神经元中的表达

摘要 目的观察δ-连环素在大鼠糖尿病模型海马神经元中的表达特征。方法根据实验需要将大鼠随机分成糖尿病组、正常对照组和缓冲剂组。按45 mg/kg一次性尾静脉注射链脲佐菌素(溶于0.1 mol/L柠檬酸盐缓冲液)诱发糖尿病,缓冲剂组只给予等量的缓冲液。应用免疫组织化学方法观察海马神经元的形态结构变化,并应用计算机图像分析技术检测δ-连环素在各组中的定量表达。结果糖尿病组大鼠海马神经元中δ-连环素表达明显低于正常对照组和缓冲剂组,在海马齿状回减少尤为明显。结论糖尿病可下调大鼠海马神经元中δ-连环素的表达。 Objective To investigate the characteristics of the expression of δ- catenin in diabetic rat hippocampal neurons. Methods The rats were randomized into diabetic, normal control and buffer groups. The rats in the diabetic group were injected with a single dose of 45 mg · kg of STZ (dissolved in 0.1 mol/L sodium citrate buffer) via the caudal vein to induce diabetes mellitus, and the animals in the buffer group received the same dose of sodium citrate buffer alone. The morphological changes of the hippocampal neurons were observed by immunohistochemical method and the quantitative expression of δ - catenin in each group was determined by computer - assisted image analysis system. Result Immunohistochemical method and quantitative assay revealed that the expression of δ - catenin in the diabetic group was the lowest in the three groups, particularly in the dentate gyms (DG). Conclusion The diabetic functional disorders of the central nervous system may downregulate the expression of δ - catenin.
