摘要
目的研究海藻提取物Fucoidan抗肿瘤作用,并对其机制进行初探。方法观察Fucoidan对小鼠S180移植性实体瘤、体外培养肿瘤细胞的作用,并了解其对动物免疫功能的影响。结果Fucoidan能明显抑制小鼠S180实体瘤的生长(P〈0.01),且呈现剂量相关性,其抑瘤率达到环磷酰胺阳性对照的70.4%-78.2%,对体外培养A549细胞、HeLa细胞和L1210细胞的生长均无显著抑制作用,Fucoidan中剂量组和大剂量组能显著提高碳粒廓清实验中的廓清指数K、吞噬指数α和脾脏器指数(P〈0.05或P〈0.01),但对胸腺脏器指数影响不大。在DNFB诱导迟发性变态反应实验中,Fucoidan中剂量组和大剂量组亦可使小鼠耳肿胀度增大。结论Fucoidan具有抗肿瘤作用,其机制可能是通过对免疫功能的增强来实现的。
Objective To observe the antitumor action of Fucoidan extracted from brown alga Undaria pinnatifida. Methods The mode of implanted S180 tumor was established in mice. The inhibitory effect on tumor was observed in mice after Fucoidan was given orally. The inhibitory effect in A549, HeLa and L1210cells by Fucoidan with the method of MTT was investigated in vitro. The immune function of Fucoidan to normal mice was detected through the experiment on the clearance rate of charcoal particles and delayed type hypersensitive reaction (DTH) induced by DNFB. Results Fucoidan showed significant inhibition on the growth of Sis0 tumor in a dose -dependent manner( P 〈 0.01 ) ,but no significant anti -proliferative effects on A.549 ,HeLa and L1210cells. The clearance rate of charcoal particles , spleen index and ear edema induced by DTH were improved in high - dose and middle - dose groups( P 〈 0.05 ; P 〈 0.01 ). Conclusion Fucoidan has antitumor action, and the mechanism maybe related to strengthening the immune function.
