河南省结核分枝杆菌临床株散在分布重复单位基因型分型法的应用

摘要 目的建立结核分枝杆菌散在分布重复单位(MIRU)基因型分型法,探讨其在河南省临床株分型应用中的可行性。方法采用简单数字表法随机抽取河南省2000年结核病流行病调查中保存的菌株149株,对其进行MIRU分型。结果MIRU分型方法可将这些菌株分为50种基因型。12个MIRU位点的多态性分析表明各位点有较大的区别,其中位点26显示了较高的多态性(h=0.69),位点31、39、40显示了中等程度的多态性(h值分别为0.36、0.30、0.32)。结论MIRU基因分型技术重复性好、快速、简便、经济,不需要昂贵的设备,是一种有效实用的结核分枝杆菌基因型分型方法。 Objective To establish a new genotyping method, mycobacterial interspersed repetitive units (MIRU) typing, and to evaluate its feasibility for Henan mycobacterium tuberculosis clinical isolates. Methods Mycobacterium tuberculosis clinical isolates were randomly selected by simple digital table from epidemiological investigation of Henan province in 2000, and were genotyped by MIRU. Results By MIRU method, the strains were divided into 50 genotypes. Twelve MIRU loci showed difference according to their allelic diversity. Locus 26 showed obvious polymorphism (h=0.69) , while locus 31, 39 and 40 showed moderate polymorphism(h=0.36, 0.30, 0.32, respectively). Conclusion MIRU genotyping is a reproducible, simple, fast and economic method, which can effectively finish genotyping for mycobaeterium tuberculosis without expensive equipments.
