摘要
研究采用常规方法提取,采用二乙酰-肟比色法,应用连续流动分析仪分光光度法测定土壤中脲酶未分解的尿素的量来估算土壤脲酶活性,避免了常规方法中重现性差,对操作者及周围环境危害较大,不易操作等缺陷。针对不同土壤类型,在东北地区采集4份混合土壤样品,测定土壤脲酶活性。结果表明,利用连续流动分析仪测定尿素的工作曲线线性较好,每个土样浸提6次,每个浸提液体连续3次平行测定的相对标准偏差均低于常规方法,即测定的精密度和重复性均高于常规方法,同时克服了测定中对人体的伤害。结果表明,采j羽连续流动分析仪结合尿素残留量法可以实现对土壤脲酶活性的精确测定。该方法简单、准确、灵敏、快速,获得了满意结果。
Improving the efficiency of Urea Fertilizer is an effective method of enhancing ineome for farmers, but it depends on measuring the soil nutrients and urease activity accurately and rapidly. Continuous-Flow Analysis method is a fast method to detect the soil nutrients. In order to evaluate the feasibility of using CFA combinated with determination of urea remainder capacity, the soil samples were collected from different types in Northeast China, and their urease activity were determined by traditional chemical method (urea remainder capacity). Then, the determination coefficient of new method was higher than traditional method, while the root mean standard error was lower. The results showed that CFA combinated with traditional method could be used to evaluate the soil urease activity accurately.
