摘要
通过对胡萝卜SRAP反应体系中Mg2+浓度、dNTPs浓度、引物浓度以及模板DNA浓度的优化,结果表明,在20μL反应体系中,DNA模板最适浓度为3.00ng/mL,Mg2+最适浓度为2.25mmoL/L,引物最适浓度为0.30μmoL/L,dNTPs最适浓度为0.20mmoL/L。
Through optimization of concentration of Mg^2＋ ,dNTPs ,primers and template DNA,SRAP-PCR of carrot was established. The results showed that in the 20μL reaction mixture the optimum concentration ofgenoinic DNA template, Mg^2＋, primers and dNTPs were 3.00ng/mL, 2.25mmoL/L, 0.30μmoL/L and 0.20mmoL/L.
