摘要
目的探讨4种微小RNA(miRNA)在前列腺癌组织中的表达及其临床意义。方法采用原位杂交(ISH)方法检测38例良性前列腺增生,52例前列腺癌及2例正常前列腺组织中4种miRNAs的表达情况。结果4种miRNAs在前列腺癌组织中的表达率与良性前列腺增生比较差异均有统计学意义(P<0.05)。4种miRNAs的表达均与肿瘤的Gleason评分相关(P<0.01);miR-15b及miR-96的表达与肿瘤的临床分期有相关性(P<0.05);miR-96的表达与肿瘤累及前列腺的叶数相关(P<0.01)。前列腺癌组织中,miR-15b与miR-96及miR-183的表达之间均存在关联性(P<0.01,r=0.41;P<0.01,r=0.45),而miR-96和miR-183的表达之间也存在关联性(P<0.01,r=0.45)。结论miRNA与前列腺癌的发生及临床生物学行为有关,并可能作为前列腺癌早期诊断及预后评估的新的生物标记物。
Objective To research the expression and their clinical significances of 4 individual microRNAs(miRNA) in prostate cancer. Methods In situ hybridization (ISH) method for the detection of 4 miRNAs was performed in 38 samples of benign prostate hyperplasia (BPH) , 52 samples of prostate cancer and 2 samples of normal prostate tissue. Results The expression of all 4 miRNAs showed a significant difference between the prostate cancer and benign prostatic hyperplasia tissues ( P 〈 0.05 ). The positive rate of these 4 miRNAs was significantly related to the Gleason Grading of prostate cancer(P 〈 0.01 ). The expression of miR-15b and miR-96 correlated with tumor clinical stage (P 〈 0. 05 ). The expression of miR-96 correlated with lobus prostatae of tumor invasion (P 〈 0. 01 ). In addition ,the expression of miR-15b was related to that of miR-96 and miR-183 ,respectively(P 〈 0. 01 ,r =0. 41 ;P 〈 0. 01 ,r =0. 45). There was a correlation between the expression of miR-96 and miR-183 in prostate cancer(P 〈 0. 01 ,r =0. 45). Conclusion The expression profiling of miRNAs may be related to the development and clinical behavior of prostate cancer, and they may be important biomarkers for early detection and prognostic assessment of prostate cancer.
