摘要
目的分别对具有原发癌特性的BxPC-3及转移癌特性的AsPC-1胰腺癌细胞株中TGFβ1及其Ⅰ型受体TGFβR1 mRNA及蛋白的表达情况进行研究。方法采用实时荧光定量RT-PCR及Western blot方法检测AsPC-1和BxPC-3胰腺癌细胞中TGFβ1和TGFβR1 mRNA及蛋白的表达。结果BxPC-3中TGFβ1和TGFβR1的mRNA和蛋白均呈低表达,AsPC-1中TGFβ1和TGFβR1的mRNA和蛋白呈高表达,两者相比较有显著统计学差异(P<0.05)。结论TGFβ1和TGFβR1的高表达可能与胰腺癌的远处转移密切相关。
Objective To compare the different expressions of transforming growth factor betal (TGFβ1) and its type one receptor(TGFβR1 ) between primary pancreatic cancer cell line AsPC-1 and metastatic pancreatic cancer cell line BxPC 3. Methods The mRNA expressions of TGFβ1 and TGF/3R1 in AsPC-1 and BxPC 3 pancreatic carcer cell lines were quantatitived by real time RT-PCR. The protein levels of TGFβ1 and TGFβR1 in these two cell lines were measured by Western blot assay. Results Compared with the primary pancreatic cancer cell AsPC-1, the mRNA and protein expressions of TGFβ1 and TGFβR1 were much higher in BxPC-3 pancreatic cancer cells(P^0. {)5). Conclusions Upregulations of TGFβ1 and TGFβR1 might be a pivotal incidence in the procedure of malignant progressing and metastasis in pancreatic cancer cells.
