IKK2dn体外转染对大鼠树突状细胞成熟度的影响 被引量:3

摘要 目的:探讨腺病毒介导IKK2dn基因(Adv-IKK2dn)体外转染大鼠树突状细胞(DCs)对其生物学行为的影响。方法:将含有IKK2dn基因的腺病毒载体转染大鼠骨髓源性DCs,获得稳定高表达IKK2dn的DCs。流式细胞术(FCM)分析转染IKK2dn基因DCs的表型变化,混合淋巴细胞反应(MLR)检测其刺激同种T细胞增殖的能力,ELISA测定MLR上清IL-10和IFN-γ水平。结果:Westernblot结果显示,转染pAdxsi-GFP-IKK2dn病毒质粒的DCs稳定表达IKK2dn。转染IKK2dn基因的DCs与未转基因的DCs相比,表面CD86和MHC-II的表达水平无显著差异。和负载BN大鼠抗原的未转染IKK2dn的DCs相比,转染IKK2dn并负载BN抗原的DCs刺激同种T细胞增殖能力显著下降(P<0.01),MLR上清中IL-10水平升高(P<0.01),而IFN-γ水平降低(P<0.01)。结论:IKK2dn基因的转染能使DCs维持于未成熟状态。同时,负载BN抗原的转IKK2dn基因的DCs可以抑制同种T细胞增殖反应,为诱导体内免疫耐受提供了实验依据。
