参芎注射液对腹膜纤维化的抑制作用 被引量:1

摘要 目的探讨参芎注射液改善腹膜纤维化的作用机制。方法将32只SD雄性大鼠随机分为4组:正常对照组(A组)、模型组(B组)、低剂量组(C组,参芎注射液15ml/kg)和高剂量组(D组,参芎注射液30ml/kg),每组8只。于实验第28天处死动物取腹膜组织待检。留取腹膜组织做HE及Masson染色,RT-PCR间接免疫荧光法检测细胞转化生长因子β1(TGF-β1)的表达。结果与A组相比,B组的壁层腹膜厚度明显增加;与B组相比,C组和D组的腹膜厚度明显减轻。B组腹膜组织TGF-β1表达明显上调,C组和D组腹膜组织TGF-β1表达明显减低。结论参芎注射液可通过抑制腹膜纤维化大鼠腹膜组织的TGF-β1表达而发挥显著的抑制纤维化的作用。 Objective To investigate the action mechanism of shenxiong injection in treating peritoneal fibrosis. Methods The model of peritoneal fibrosis was induced by high glucose dialysate (4. 25 %, Baster) and lipopolysaccharide (LPS). The rats were randomly divided into 4 groups: control group, model group, low dose shenxiong injection-treated group and high dose shenxiong injection-treated group The shenxiong injection-treated groups received daily intraperitoneal injection of 15 and 30 ml/kg shenxiong injection respectively. All rats were sacrificed after 4 weeks. The abdominal membrane was stained with HE and Masson methods. The expression of TGF-β1 in peritoneal membrane was detected by using immuno-fluorescent and RT-PCR methods. Results The peritoneal membrane in model group was significantly thickened. As compared with model group, the peritoneal membrane in shenxiong injection-treated groups was thinned markedly. The expression of TGF-β1 was obviously up-regulated in model group as compared with model group, and that in low dose and high dose shenxiong injection-treated groups was notable decreased. Conclusions shenxiong injection plays a significant role in anti-fibrosis by inhibiting the expression of peritoneal TGF-β1.
