摘要
目的:研究三七总皂苷(panax notoginseng saponins,PNS)对佐剂性关节炎(rheumatoidarthritis,AA)大鼠的抗炎及免疫调节作用。方法:大鼠右后足跖皮内注射弗氏完全佐剂诱发大鼠AA模型,进行多发性关节炎评分,放射免疫法检测大鼠血清肿瘤坏死因子(tumor necrosis factor,TNF)、白细胞介素-1β(interleukin-1β,IL-1β)含量,同时对脂多糖(lipopolysaccharide,LPS)诱导的AA大鼠腹腔巨噬细胞(peritoneal macrophages,PM)产生的TNF、IL-1β进行检测。结果:大鼠免疫后2周,PNS腹腔注射给药2周,可降低AA大鼠多发性关节炎指数(P<0.05),抑制血清TNF(P<0.01)、IL-1β(P<0.05)产生;PNS体外分别给药0.8,0.4,0.2 mg/ml,作用24 h,可不同程度抑制LPS诱导的AA大鼠PM产生TNF(P<0.05,P<0.01)和及IL-1β(P<0.05)。结论:PNS可能通过抗炎及抑制血清、PM释放炎性细胞因子TNF、IL-1β对AA大鼠发挥治疗作用。
Objective:To investigate the anti-inflammatory and immunomodulation effects of panax notoginseng saponins(PNS) in rats with adjuvant arthritis (AA). Methods: Complete Freund's adjuvant was used to induce AA in rats. Polyarthritis index were scored and levels of tumour necrosis factor-α (TNF-α) and interleukin-1β(IL-1β)from serum were determined by radio immunoassay. Meanwhile, levels of TNF-α and IL-1β produced by I.PS induced PM of AA rats were also determined by radio immunoassay. Results: PNS were given by intraperitoneal injection for 2 weeks from the 15th day after immunization . The polyarthritic symptoms of AA rats were improved significantly compared with those of AA model group(P〈 0.05). PNS significantly reduced the enhanced productions of TNF-α(P〈0.01) and IL-1β(P〈0.05)from serum in AA rats. Meanwhile, PNS significantly reduced the enhanced productions of TNF-α(P〈0.05, or P〈0.01) and IL-1β(P〈0.05)produced by LPS-indueed PM of AA rats at different concentration (0. 8, 0.4 and 0.2 mg/ml, respedively). Conclusion:PNS has therapeutic effect on AA in rats, which may be related to anti-inflammatory and immunomodulation effects of inhibiting productions of proinflammatory cytokines of TNF-α and IL-1β from serum and LPS-induced PM.
