摘要
目的:建立板蓝根中抗病毒有效成分clemastanin B的高效液相色谱测定方法,测定不同产地板蓝根中clemas-tanin B的含量。方法:采用反相高效液相色谱法,色谱柱为ZORBAX SB-C18(4.6 mm×250 mm,5μm) 流动相为乙腈-水11∶89 流速为1.0 mL.min-1 检测波长为225 nm,柱温为30℃。结果:clemastanin B在0.061 5~1.844 1μg与峰面积线性关系良好(r=0.999 5),平均加样回收率为97.74%,RSD 1.4% 测得8批板蓝根中clemastanin B的质量分数为0.269~0.900 mg.g-1,其中以河北安国产板蓝根药材中clemastanin B含量最高,安徽阜阳、河北承德和安徽亳州产的次之,其他几个产地的含量相对略低,甘肃岷县产的最低。结论:该方法准确、可靠,可用于板蓝根药材的测定。
Objective: To develop an HPLC method for determination of clemastanin B which has anti-viral activity in Radix Isatidis and compare the contents of elemastanin B in the drugs from different origins. Method: The samples were separated on an ZORBAX SB-CI8 ( 4. 6 mm ~ 250 mm, 5 p,m) column with the mobile phase of acetonitrile-water ( 11 : 89 ). Flow rate was 1.0 mL ~ min-1. The detection wavelength was set at 225 nm. Column temperature was 30 ℃. Result: The linear range of clemastanin B was 0. 061 5-1. 844 1 p,g (r =0. 999 5), the average recovery was 97. 74%, RSD was 1.4% (it =9). The contents of clemastanin B were in the range of 0. 269-0. 900 mg · g-1 in Radix Isatidis from different origins. Conclusion: The method for quantitation of elemastanin B in Radix Isatidis was accurate and reliable, which can be used to evaluate the quality of Radix Isatidis.
