人血清脂联素定量ELISA检测方法的建立及初步临床应用 被引量:3

摘要 目的:建立一种检测人血清中脂联素(APN)的酶联免疫吸附方法(ELISA),并初步探讨其在冠心病诊断中的应用。方法:构建表达质粒pET22b(+)/gAPN,在原核表达系统大肠杆菌中表达并纯化重组脂联素球状区(gAPN)蛋白。用抗APN单克隆抗体MAB10651包被微孔板,10%小牛血清作为封闭液,针对APN不同抗原表位的单克隆抗体MAB1065生物素化后作为标记抗体,联合生物素-亲和素信号放大系统,以gAPN重组蛋白为标准品绘制标准曲线,以准确性、特异性、重复性、回收率试验和方法对比试验评价ELISA方法。检测161例冠心病患者血清APN水平并结合冠状动脉造影结果和Gensini积分系统,探讨血清APN水平与冠心病的关系。结果:利用基因克隆技术成功构建了pET22b(+)/gAPN质粒,获得了相对分子质量约15000的重组gAPN蛋白,产量较高(1.57mg),纯度>90%,并以此为标准品初步建立了检测人血清APN的定量ELISA方法。该法具有良好的准确性和重复性,灵敏度达150pg/ml,回收率为91.0%~108.0%,与深圳依诺金生物科技有限公司进口分装的ELISA试剂盒对照比较有良好的相关性(r=0.935,P<0.001)。临床检测表明冠心病组血清APN水平明显低于非冠心病组(P<0.01),且血清APN水平随冠状动脉狭窄程度的加重呈进行性下降趋势。结论:本实验成功建立了一种简便快速检测人血清APN的ELISA方法,对诊断冠心病有临床应用价值,为国内APN诊断试剂盒的研制和开发提供了科学实验依据。
