摘要
目的观察碱性成纤维细胞生长因子(bFGF)对体外培养人乳腺癌MCF-7细胞增殖和凋亡的影响,初步探讨bFGF作用机制。方法在饥饿培养的MCF-7细胞中加入bFGF和PD98059处理,以MTT法、吖啶橙染色及流式细胞术观察细胞生长与凋亡情况;并用Western blot检测caspase-3蛋白含量。结果对照组细胞形态发生改变:核质固缩、有凋亡小体形成;细胞凋亡率较高;Western blot分析表明,caspase-3蛋白明显表达。bFGF处理后,细胞变饱满,凋亡现象减少;细胞增殖比明显增加;与对照组相比凋亡细胞比例下降,并诱导细胞进入S期;随着bFGF浓度增加,caspase-3蛋白表达水平降低,在一定范围内呈剂量依赖性。加入PD98059可抑制bFGF的这些作用。结论bFGF可以促进细胞增殖,加速人乳腺癌细胞系MCF-7细胞的细胞周期进程,抵抗无血清饥饿诱导的凋亡,其作用部分可能是通过Ras-Raf-ERK1/2途径介导的。
Objective To investigate the effect of basic fibroblast growth factor (bF'GF) on the proliferation and apoptosis of human breast cancer cell line MCF-7, and to study the function mechanism of bFGF. Methods Induced by starvation, the MCF-7 cell line was treated with hFGF and PD98059. The cell proliferation and apoptosis were analyzed by MTT assay, acridine orange (AO) dyeing, flow cytometry, respectively. The expression of caspase III was determined by Western blot. Results Apoptotic morphology of the control group was observed: the nuclear shrinkage and higher apoptotic rate. Western blot showed remarkable caspase III expression. Treated with bFGF, the cell morphology was changed: the proliferation of MCF-7 cell line increased; the apoptotic rate was significantly lower than that in the control group, and S-phase was induced, bFGF inhibited caspase III activity in a dose-dependent manner. All above effects were inhibited by PD98059. Conclusion In human breast cancer cell line MCF-7, bFGF increase the prolif- eration rate, promotes the cell cycle, and decrease the apoptotic rate via the Ras-Raf-ERK1/2 pathway.
