摘要 为测定麦角固醇和维生素D2及其他光化学异构体,研究确定了HPLC—ELSD测定的色谱分离条件:KromasilC18柱(4.6×250mm,5μm),流动相为体积比95:5甲醇-水溶液,流速1.3ml/min,蒸发光散射检测器漂移管温度为75℃,气体流量1.6L/min。在此条件下,各组分得到有效地分离,麦角固醇和维生素D2色谱峰面积的自然对数与浓度的自然对数呈良好的线性关系(r〉0.997),精密度RSD为1.89%~2.75%,最低检测限为0.05--0.1μg,回收率测定值为101.4%~102.8%。 An I-IPLC-ELSD method for the determination of ergosterol and the photochemical iso- mers of vitamin I32 were studied. Chromatographic analysis was carried out on a Kromasil C18 column(4.6 mm×250mm,5μm) with methanol-water (95:5,V/V)as the mobile phase. The flow rate was 1.3 ml/min. The drift tube temperature was at 75℃ and gas flow rate was 1. 6SLPM. The relative standard deviation (RSD) was below 2.75 % ( n = 6), and detection limits attainable were in the range 0.05μg for ergosterol to 0.1μg for vitamin D2, and the recoveries were from 101.4 % to 102.8 %.
