大鼠心肌缺血后心、肝组织应激改变的研究 被引量:2

摘要 目的本研究旨在探讨短暂和持续心肌缺血应激后大鼠心、肝组织病理改变及热休克蛋白(HSP70)的表达规律。方法复制大鼠短暂心肌缺血(BMI)5min后再灌20h及持续心肌缺血(MI)20h模型,取心、肝组织,用Western blotting法、免疫组化及HE染色,观察大鼠心、肝组织的病理变化和HSP70的表达规律。结果短暂和持续心肌缺血后心、肝组织病理形态均有改变,HSP70表达均显著增加,心、肝组织中HSP70表达在短暂心肌缺血再灌组增加显著且组织病变较轻,持续心肌缺血组HSP70表达较少且组织损伤较大。结论不同程度心肌缺血对心、肝组织病理改变和HSP70表达影响不同,短暂心肌缺血再灌比持续心肌缺血对心脏及肝脏组织损伤较小而刺激组织HSP70增加较多。 Objective To investigate the pathological changes and the expression rule of heat shock protein production(HSP70) in heart and liver tissues of rats after brief myocardial ischemia (BMI) and persistent myocardial ischemia(PMI). Methods The models of BMI and PMI were made in rats. 24 healthy male SD rats were randomly divided into 4 groups: control group( n = 6) , persistent myocardial ischemia (PMI) group ( n = 6), brief myocardial ischemia-reperfusion (BMI) group ( n = 6), sham ligation group ( n = 6). At 20 hours after operation, the rats were killed, and the pathological changes in heart and liver tissues of rats were detected by immunohistochemistry stain and HE stain. The expression rule of HSP70 was observed by Western Blot. Results The pathological changes in heart and liver tissues of rats could be foun after BMI and PMl,and the expression of HSP70 was obviously increased, the expression of HSP70 in heart and liver tissues in BMI group was increased significantly and the pathological changes were relatively gentler, as compared with that in PMI group, however, the expression of HSP70 in PMI group was relatively less and the damage of tissues was comparatively severe. Conclusion The influence of different degrees of myocardial ischemia on the pathological changes and the expression of HSP70 is quite different, and the injury effect of BMI on heart and liver is less than that of PMI, while BMI can stimulate tissues to increase the expression of HSP70.
