摘要
为探讨氧化应激在硒缺乏致鸡淋巴细胞DNA损伤中的作用,选择1日龄健康海兰褐雏鸡120只,随机分为对照组和缺硒组,对照组饲喂硒含量为0.282mg/kg的饲料,缺硒组饲喂硒含量为0.032mg/kg的饲料,分别于30、45、60及75日龄采集胸腺、脾脏和法氏囊,检测硒含量、抗氧化功能和淋巴细胞DNA损伤。与对照组比较,缺硒组鸡胸腺、脾脏和法氏囊中硒含量降低;T-AOC,GSH-Px和SOD活性降低,MDA含量升高;淋巴细胞彗星率增加,并呈明显的时间-效应关系;胸腺、脾脏和法氏囊中硒含量、T-AOC、GSH-Px和SOD活性与淋巴细胞DNA损伤呈负相关,MDA含量与其呈正相关。结果表明,氧化应激与硒缺乏致鸡淋巴细胞DNA损伤密切相关。
To study the effect of oxidative stress on lymphocyte DNA damage induced by selenium deficiency in chickens,120 1-day-age healthy Hy-Line brown chickens were randomly divided into control group and selenium deficiency group. The feed contained 0.282 mg/kg Se in the control group while the selenium deficiency group was fed the diet with 0.032 mg/kg Se. Thymus,spleen and bursa of fabricius were collected and the selenium content,oxidation resistance and lymphocyte DNA damage were detected after 30,45,60 and 75 days age. Compared with the control group,the selenium content decreased,T-AOC,SOD and GSH-Px activity in selenium deficiency group reduced and MDA content increased. The comet rate of lymphocyte increased,accompanied with evident time--effect relationship. The selenium content,T-AOC, SOD and GSH-Px activity were negative correlation with DNA damage of lymphocyte,a significant positive correlation was found between the MDA content and lymphocyte DNA damage. It has close relationship between oxidative stress and lymphocyte DNA damage induced by chickens with selenium deficiency.
