摘要
本研究对已整合外源rolB-pttGA20ox双价基因的毛白杨株系进行半定量RT-PCR检测,并对移栽后的转基因植株进行形态观测。结果表明:rolB和pttGA20ox基因的表达模式不同,rolB基因在不同器官中的表达强度为根>茎>叶,pttGA20ox基因的表达无组织特异性;rolB基因受外源生长素短时间诱导时表达丰度发生变化,pttGA20ox基因表达丰度不受外源生长素短时间诱导。对移栽温室后的转化植株的生长量分析结果表明,4个转化株系及对照的高生长增加量表现为RG-4>RG-1>RG-3>RG-2>CK。4个转化株系的株高和茎间长明显高于对照。转化株系与对照之间以及转化植株之间的形态特征出现了差异。
In this study, the expression of rolB - pttGA20ox double genes in transgenic lines of Populus tomentosa was analyzed by RT - PCR. The results showed the differences between rolB and pttGA20ox gene. The expression levels of rolB gene in different tissues were root 〉 stem 〉 leaf, and pttGA20ox gene had same expression levels in these tissues. The rolB message level was changed among exposure to the auxin treatment in 20 h and pttGA20ox gene wasn't induced by auxin treatment. Growth analysis showed that the increases of growth were RG- 4 〉 RG - 1 〉 RG - 3 〉 RG- 2 〉 CK. The plant height and internode length of four transgenic lines were significantly higher than those of the control. The transgenic plants differed in morphology among themselves and from the control plants.
