脑出血患者血管紧张素原T704C基因多态性分析

Analysis of the relationship between intracerebral hemorrhage and angiotensinogen T704C gene polymorphisms.

摘要 目的:探讨脑出血(ICH)患者血管紧张素原(AGT)T704C基因多态性及二者的相关性。方法:采用病例—对照研究,结合聚合酶链反应—限制性片断长度多态性(PCR-RFLP)技术检测70例ICH患者和89例健康体检者的AGT T704C单核苷酸多态性,并将ICH组按是否合并高血压而分为两组:高血压组和非高血压组。比较和分析ICH与对照组间该基因型及等位基因的分布频率差异,并分析该遗传因素对ICH的作用。结果:ICH组704CC基因型频率41.4%,C等位基因频率67.1%,与对照组比较,差异无统计学意义(P>0.05)。结论:AGT T704C基因多态性可能与ICH的发生无关。 Objective: To study the distributive characters of T704C of angiotensinogen(AGT)gene's single nucleotide polymorphisms(SNP), and to investigate the association between AGT gene SNP and intracerebral hemorrhage(ICH). Methods: A case-control study of 70 cases and 89 controls were carried out with polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP). The AGT genotype and gene frequencies were analyzed, and further analysis were conducted according as they were hypertensive or without hypertension. Distributive differences of AGT genotype and allele frequencies were analyzed in the case group and control group, then concluded whether this hereditary factor could lead to ICH. Results:Three genotypes in site 704 of AGT gene were identified among 169 individuals. The CC genotype's frequency of AGT gene was 0. 414 in cases and 0. 348 in controls(P〉0.05);The C allele frequency of AGT gene was 0. 671 in cases and 0. 612 in controls(P〉0.05) ; and there were no significant difference in the frequencies of CC genotypes and C allele between the two subgroups of ICH(P〉0. 05). Conclusions: AGT T704C SNP existed in our population group and AGT T704C SNP was not associated with the occurance of ICH.
