巯基化羧甲基壳聚糖微凝胶的制备及其生物黏附性能 被引量:7

摘要 巯基化羧甲基壳聚糖是羧甲基壳聚糖的衍生物。它既保留了羧甲基壳聚糖的优良生物特性,具备水溶性和pH敏感性,同时也具备了优良的生物黏附性,是制备生物凝胶材料的优良原料。利用L-半胱氨酸进行巯基反应制备巯基化羧甲基壳聚糖,通过1-乙基-3-(3-二甲胺基丙基)-碳二亚胺盐酸盐交联和自体交联等方式制备了微凝胶,并以猪胃黏蛋白为模型,测定微凝胶的体外生物黏附性。实验表明巯基化羧甲基壳聚糖粉末在pH3.0的介质中有最大黏附率(120.25%),表现出了优良的生物黏附性能,在相同的介质中,自身交联的巯基化羧甲基壳聚糖微凝胶的黏附能力要大于1-乙基-3-(3-二甲胺基丙基)-碳二亚胺盐酸盐交联的微凝胶。 Thiolated carboxymethyl chitosan (SH-CMC), the derivative of carboxymethyl chitosan (CMC), is a water-soluble and pH sensitive material, while retained the excellent bioadhesion, so it is a superexcellent material to prepare biogelatin. L-cysteine was used to prepare the SH-CMC. Micro-gel based on SH-carboxymethyl chitosan was prepared by crosslinking with 1-ethyl-3-(3-dimethylamino-propyl)-carbodiimide hydrochloride (EDC) and itself. The in vitro biological adhesion of micro-gel was determined, and pig gastric mucin was used as a model protein. Experiments showed that SH-carboxymethyl chitosan the biological adhesion of micro-gel had the biggest rate of 120.25% in pH3.0. In the same medium, the adhesion ability of micro-SH CMC gel crosslinked by itself is greater than EDC crosslinking.
