摘要 目的:探讨TNF-α作用于人乳腺癌MCF-7细胞后对CD44蛋白表达和细胞迁移的影响及其可能的机制。方法:细胞迁移率实验检测TNF-α作用后MCF-7细胞迁移率的变化;Western blotting方法检测TNF-α作用后CD44s、CD44v3和CD44v6蛋白表达水平的变化,以及MAPK信号传导通路蛋白p38、JNK、ERK表达水平的变化。结果:经TNF-α作用后,MCF-7细胞迁移率提高,细胞CD44s蛋白表达水平下降,CD44v3、CD44v6蛋白表达上升;TNF-α作用后可以激活JNK信号途径,对p38和ERK信号通路无明显效应。结论:TNF-α可能通过激活JNK信号传导途径调控CD44各亚型蛋白表达,进而影响人乳腺癌细胞MCF-7的迁移能力。 Objective:To detect the effect of tumor necrosis factor-α on CD44 expression and cell migration in human breast cancer cell lines MCF-7, and explore the probably mechanisms during the process. Methods:MCF-7 cells were treated by various concentrations of TNF-α for 24 hours. Migration capability was detected using cell migration assay comparing the cells treated with 20ng/ ml TNF-α with the untreated ones. The expression of CD44s, CD44v3 and CD44v6 protein were measured by Western blotting analysis. In addition, the effect of TNF-α on MAPK( mitogen-activated protein kinase) signaling transduction pathways, including p38, JNK and ERK pathways, were examed after TNF-α treated for various times. Results:After treatment of TNF-α, the cell migration capability was enhanced. The expression of CD44s was decreased, while a visible increase in CD44v3 or CD44v6 was detected. TNF-α could activate the JNK pathway, but have no obviously effect on p38 or ERK pathways. Conclusion:TNF-α may regulate the expression of CD44 through activating the JNK pathways, and in advance affect the migration capability of human breast cancer cell MCF-7.
