缺氧刺激后大鼠大脑皮层神经元GABA和ACh表达的动态变化 被引量:4

摘要 目的研究大脑皮层神经元中枢神经疲劳过程中γ-氨基丁酸(GABA)和乙酰胆碱(ACh)神经递质表达的变化及其特点。方法借助体外培养大鼠大脑皮层神经元,并利用Western Blotting、HPLC等方法研究60 min缺氧刺激对神经元HIF-1α、GABA和ACh表达的影响。结果缺氧刺激后HIF-1α、GABA和ACh的表达均有显著变化,两种递质的变化呈现异向同步性;24-48 h后,二者的变化基本恢复正常。结论缺氧可以诱导大鼠大脑皮层神经元GA-BA和ACh的表达升高和降低,二者协同作用可以诱导神经元的自我保护效应。 Objective The purpose of this study is to explore the dynamic change characteristics of of gamma - aminobutyric acid (GABA) and acetylcholine (ACh) induced by hypoxic stimulus presented in neuron of central nervous system under mimetic fatigue condition. Methods The in vitro culturing neuron derived from cerebral cortex of mice and the methods of RT - PCR, Western Blotting and high performance liquid chromatography (HPLC) were used to inspect the intracellular expression of hypoxia- induced factor - alpha ( HIF - 1a), GABA and ACh intervend by 60 - min hypoxic stimulus. Results The hypoxic stimulus significantly increased the expression of GABA and decreased the expression of ACh in telencephalic neuron, with allotridirection synchronism tendency, which followed behind the significant expression of HIF- 1a. Within 24h- 48h, those hypoxic contributions could vanish. Conclusion Hypoxia could induce the activation of self- protection of telencephalic neuron of mice, which was mediated by the synergetic mechanism induced by increased expression of GABA and decreased expression of ACh.
