羌活醇提物预处理对大鼠HIRI模型的防护作用 被引量:3

摘要 目的探讨羌活醇提物对大鼠肝缺血再灌注损伤(hepatic ischemia reperfusion injury,HIRI)模型肝功能和抗氧化酶的影响。方法将60只Wistar大鼠分为空白组(6只)、模型组(18只)、丹参预处理组(DS组,18只)和羌活预处理组(QH组,18只),后三组组内再分为缺血45min组(I45)、缺血45min再灌注30min组(I45R30)和缺血45min再灌注60min组(I45R60)3小组,每组6只,造模后测定大鼠各时点即刻的血清丙氨酸氨基转移酶(alanine amiotransferase,ALT)、门冬氨酸氨基转移酶(aspartate aminotransferase,AST)水平以及大鼠肝组织中丙二醛(malondiadehyde,MDA)含量、超氧化物歧化酶(superoxide dismutase,SOD)活力水平。结果MDA:羌活组I45R30min即刻点MDA含量明显高于I45min即刻点(P<0.01);I45min即刻点,羌活组MDA含量高于模型组(P<0.05);I45R30min即刻点,丹参组与羌活组MDA含量均明显高于模型组(P均<0.01);SOD:羌活组I45R30min和I45R60min时点的肝组织SOD活力均高于I45min时点(P均<0.01);I45R30min即刻点模型组肝组织SOD活力均低于丹参组和羌活组(P均<0.001)。结论羌活醇提物可能是通过提高HIRI大鼠肝组织的总SOD活力,增强其对氧自由基的清除能力,减轻脂质过氧化损伤,从而起到保护肝脏的作用。 Objective To study the effect of ethanol soluble extract of Notopterygium incisum on liver function and antioxidase in hepatic ischemia -reperfusion injury model of rats. Methods 60 healthy adult wistar rats were randomly divided into four groups: sham group (Sham, 6), model group (Model, 18), Danshen pretreatment group (DS, 18) and Notopterygium incisum pretreatment group(QH, 18). The latter three groups subdivided into three groups respectively, namely, ischemia 45 min subgroup (I45, 6 ), ischemia reperfusion 30 min subgroup ( I45R30, 6) and ischemia reperfusion 60 rain subgroup ( I45R60, 6). Rats from each group were sacrificed to determine serum ALT (alanine amiotransferase, ALT), AST (aspartate aminotransferase, AST) and the activity of MDA (malondialdehyde,MDA) and SOD (superoxide dismutase, SOD). Results (1)MDA: In the QH group, MDA in I45R30 subgroup was significantly higher than I45 ( P 〈 0.01 ). In the I45 subgroup, the QH group washigher than the model group(P 〈0.05). In the I45R30 subgroup, the MDA in DS and QH group were higher than the model group (P 〈0.01). (2) SOD: In the QH group, SOD activity in the I45R30, I45R60 subgroup were significantly higher than I45 ( P 〈 0.01 ). In the I45R30 subgroup, the SOD activity of model group was significantly reduced than the DS group( P 〈0.01 ) and QH group( P 〈 0. 001 ). Conclusion The ethanol soluble extract of Notopterygium incisum may promote activity of T - SOD in the liver tissue, strengthen the ability of cleaning significantly by both enzymes mentioned above and reduce the lipid peroxidation injury. In summary, Notopterygium incisum show a preventive effect on hepatic ischemia reperfusion injury in rats.
