摘要
目的研究乳腺癌中整合素β3mRNA的表达及其与肿瘤血管生成、生长方式、浸润转移和预后的关系。方法应用原位杂交和免疫组织化学S-P法检测102例乳腺癌中整合素β3mRNA和CD34的表达。结果整合素β3mRNA在乳腺癌中阳性表达率为48%,在乳腺癌中有腋窝淋巴结转移组61.8%,无腋窝淋巴结转移组31.9%,两者差别有统计学意义(P<0.05)。CD34阳性血管数作为MVD。整合素β3mRNA阴性表达组的MVD值为(17.35±5.67),整合素β3mRNA阳性表达组的MVD值为(20.37±7.30),两者差别有统计学意义(P<0.05);CD34阴性表达组的MVD值为(17.14±5.78),CD34阳性表达组的MVD值为(21.27±7.14),两者差别有统计学意义(P<0.05)。结论浸润性癌中的整合素β3mRNA的表达与腋淋巴结转移成正相关,与患者的年龄、肿瘤大小、ER、PR无相关性;在乳腺癌组织中MVD表达与整合素β3mRNA表达成正相关。
Objective To study the expression of integrin beta-3mRNA in breast cancer and to find the correlation between the expression and vascularization of tumor,growth pattern,metastasis of infiltration,prognosis. Methods In situate hybridization and immunohistochemical s-p techniques were used to study the gene expression of Integrin beta-3 mRNA and CD34 in 102 breast cancer specimens. Results In situ hybridization revealed that positive expression rate of Integrin beta-3 mRNA was 48% in breast cancer cases,was 61.8% in axillary nodes metastasis group and was 31.9% in control group,the difference has statistical significance(P〈 0.05);Mean tumor MVD of patients with negative beta-3 mRNA expression was( 17.35 ± 5.67);the value of patients with positive beta-3 mRNA expression was(20.37 ± 7.30), they have significancent difference(P〈 0.05). Mean tumor MVD of cases with negative CD34 expression was( 17.14 ± 5.78);the value of cases with positive CD34 expression was(21.27 ± 7.14), they have significancent difference(P 〈 0.05 ). Conclusion the expression level of Integrin beta-3 mRNA was positively correlated with axillary nodes metastasis in invasive breast cancer(P 〈 0.05), and had no correlation with age, size of tumor, ER, PR; The expression level of MVD was positively correlated with the expression level of Integrin beta-3 mRNA in tissue of breast cancer.
