摘要
目的探讨CD40配体激活对肺癌细胞生长、增殖的影响。方法采用CCK-8法检测不同浓度sCD40L作用后A549肺癌细胞株存活率,选定最佳干预浓度;流式细胞术检测sCD40L最佳干预浓度作用后肺癌细胞株A549、SPC-A-1细胞周期变化。结果sCD40L的最佳干预浓度为2μg/ml,与肺癌细胞株A549、SPC-A-1共培养48 h,sCD40L可使高表达CD40的A549细胞生长抑制,细胞周期阻滞在G1期,而对CD40低表达的SPC-A-1细胞生长无明显影响。结论sCD40L通过阻滞细胞周期可明显抑制CD40高表达肺癌细胞的生长。
Objective To observe the effect s of CD40 ligand activations on the growth and proliferation of lung cancer cell lines. Methods The growth and proliferation of lung cancer cell lines with different CD40 ligand concentration were first measured by eek-8, then the best CD40 ligand activating concentration were selected. Cell cycle of A549 ,SPC-A1 lung cancer cell lines activating with the best concentration were analyzed by flow eytometry. Results The best CD40 ligand activation concentration was 2 μg/ ml. Inhibition of growth and blockage of cell cycle were observed in A549 cells expressing high levels of CD40 after CD40 ligand activating 48 hours. But SPC-A1 cells without expressing CD40 didn' t respond to it. Conclusion The growth of lung cancer cells expressing high level of CD40 is suppressed by CD40 ligand activation. And the mechanism may be to block cell cycle.
