摘要
目的探讨PTEN、MDM2和c-erbB-2基因在膀胱移行细胞癌发生发展中的临床意义。方法采用RT-PCR技术,检测32例膀胱移行细胞癌癌组织、癌旁组织及正常膀胱组织中PTEN、MDM2、c-erbB-2基因的表达。结果PTEN、MDM2和c-erbB-2在膀胱癌组织中阳性表达率分别为31.3%、50.0%、78.1%(P<0.05)。PTEN在正常组织中呈高表达,随肿瘤分级、分期的增加呈递减趋势(P=0.002)。MDM2在正常组织中表达率是3.1%,而在G1级膀胱癌组织中呈高表达,并且随肿瘤分级、分期的增加呈递减趋势(P<0.01)。c-erbB-2在正常组织中不表达,而在肿瘤组织中呈高表达,并且随肿瘤分级、分期的增加呈递增趋势(P<0.01)。PTEN与MDM2表达、c-erbB-2表达均呈负相关(γ=-0.572,P=0.015;γ=-0.653,P=0.025),MDM2与c-erbB-2无相关性(P>0.05)。结论PTEN、MDM2和c-erbB-2基因的表达异常及协同作用在膀胱移行细胞癌的发生和发展过程中起重要作用。
Objective To investigate the genes expression of PTEN, MDM2 and c-erbB-2 in transitional cell carcinoma of bladder(BTCC) and its significance. Methods PTEN, MDM2 and c-erbB-2 genes were examined in the tissues from BTCC, adjacent and normal tissue by using reverse transcfiptase polymerase chain reaction (RT-PCR) assay. The correlation between these gene expressions and the biological behaviors of BTCC were also evaluated. Results The expression of PTEN, MDM2 and c-erbB-2 mRNA in BTCC tissue were 31.3 % ,50.0% , 78.1% ( P 〈 0.05 ) , respectively. PTEN, MDM2 expression was declined with the increasing tumor grade and stage ( P 〈 0.05 ), and e-erbB-2 expression was increased ( P = 0. 002 ). A significantly negative correlation was observed between the expression of PTEN and MDM2 ( γ = -0. 572, P = 0. 015 ) , and c-erbB-2 ( γ = - 0. 653, P = 0. 025 ). No correlated was seen between the expression of MDM2 and c-erbB-2. Conclusion It is suggested that ab- normal expression and synergism of PTEN, MDM2 and c-erbB-2 mRNA may play an important role in the genesis and malignant progression of BTCC.
