大鼠全脑缺血再灌注损伤时海马细胞SOD、MDA及Cdk-5的变化

摘要 目的:探讨急性全脑缺血再灌注损伤时大鼠海马超氧化物歧化酶(superoxidismutase,SOD),丙二醛(Malondialdehyde,MDA),细胞周期依赖性蛋白激酶-5(cyclin-dependent kinase 5,Cdk-5)的关系变化。方法:用三管结扎法建立急性全脑缺血再灌注大鼠模型,用免疫沉淀法测定Cdk-5的酶活性,用比色法测定SOD的活性及MDA的含量。结果:与对照组比较,在缺血和/或再灌注大鼠海马神经元内SOD活性降低(P<0.01),MDA含量明显升高(P<0.01),且伴有细胞内Cdk-5活性升高(P<0.05,P<0.01)。结论:本研究从整体水平上证实急性全脑缺血-再灌注时大鼠海马神经元Cdk-5活性升高,与氧化应激反应密切相关。 Objective:To explore the corralation of superoxidismutase, malondialdehyde and cyctin-dependent kinase 5(Cdk-5) in the hippocampus of global ischemia-reperfurion rat brains,Methods:A model of brain ischemia-reperfusion was established by clipped three arteries and then released to reperfuse blood into brain in rats.The rats were divided randomly into control,ischemia and ischemia- reperfurion(I-R)groups.SOD activity and MDA contain were detected by colorimetry, and the activity of Cdk-5 was measured by immtmoprecitation. Results:Compared with control group,SOD activity(P〈0.01,P〈0.01)was decreased and MDA contain was increased (P〈0.01,P〈0.01)in iscncmia group and in ischemia-reperfurion group(P〈0.0i),and an etevated activity of Cdk-5 was inducedin ischemia group(P〈0.05)and in ischemia-reperfurion group(P〈0.01).Conclusions:The higher level of Cdk-5 activity was tightly correlative with oxidative stress in the hippocampus of global ischemia-reperfurion rat brains.
