抗坏血酸对大鼠神经干细胞分化影响的体外实验探讨

摘要 目的:探讨药物抗坏血酸对神经干细胞(NSCs)体外分化的作用。方法:体外培养和鉴定NSCs,细胞免疫学方法检测分化细胞中酪氨酸羟化酶(TH)表达。结果:对照组、AA20μm、AA50μm、AA100μm、AA150μm诱导7d后TH阳性细胞分别平均为1.5%、3.2%、5.5%、5.7%。结果显示AA处理后NSCs分化细胞明显TH阳性细胞表达率升高,与对照组比较有显著性差异(P<0.05)。结论:抗坏血酸AA体外对神经干细胞分化过程有重要的影响作用。 Objective:To explore the effects of AA on differentiation into neurons from NSCs in vitro. Methods: Culture and identify neural stem cells in vitro.We use immunocytochernistry to detect TH-positive neurons induced by the control and AA for 7days respectively.Results:The positive ratios of TH expression in induced cells by the control,AA20μm,AA50μm,AA100μm,AA150μm were 0.45%,1.5%,3.2%,5.5%,5.7% respectively.The positive ratios of TH expression in induced ceils by AA were more higher than by the control(P〈0.05).Conclusion:The induction of AA on differentiation into neurons from NSCs in vitro is significant.
