摘要
目的:研究层粘连蛋白(laminin,LN)及其受体(lamininreceptor,LNR)在早、中、晚期人胎盘绒毛的定位及其相对含量变化。方法:取早期、中期和晚期胎盘绒毛,常规制作石蜡组织切片,采用免疫组织化学ABC法和图像分析技术进行LN及LNR定位和定量测定。结果:LN阳性反应主要位于绒毛上皮基膜和绒毛血管内皮基膜;LNR的阳性反应位于绒毛上皮合体滋养层表面及合体滋养层和细胞滋养层的细胞质内,细胞核呈阴性。LN及LNR的含量以早期胎盘绒毛中最高,中期及晚期含量明显降低。结论:人胎盘绒毛含有LN及LNR,随着胎盘周龄的增加。
ABSTRACT Objective: To study the localization and quantification of laminin (LN) and laminin receptor (LNR) in the human placental villi during early, middle and term pregnancy. Methods: LN and LNR in the human placental villi was measured using immunohistochemical ABC method and Q500 image analysis system. Results: LN positive immunostaining was found in placental villous epithelial basement membranes and vascular endothelial basement membranes. LNR positive immunoreactive substance was localized in the villous epithelial cellular plasma (including cytotrophoblast and syncytiotrophoblast ) and syncytiotrophoblastic cellular surfaces, but not in the nucleus. Along with placental development, the amount of LN and LNR decreased gradually. Conclusion: LN and LNR exist in the human placental villi, and their relative content decreased following the placental growth.
