调肝脾方与补肝肾方对佐剂大鼠红细胞免疫和细胞因子影响的比较研究

Comparative study on adjusting liver and spleen prescription and tonifying liver and kidney prescription on erythrocyte immune and cytokines of adjuvant rats

目的:比较调肝脾方和补肝肾方对佐剂大鼠血清细胞因子白细胞介素-4(IL-4)、干扰素-γ(IFN-γ)及其比值以及红细胞免疫功能的影响。方法:Wistar大鼠按体质量随机分为6组,即正常组、模型组、调肝脾方药组、补肝肾方药组、火把根片组、芬必得组,采用佐剂致炎方法造成免疫性关节炎大鼠模型,并灌胃给服不同药剂。采用酶标法检测血清IL-4、IFN-γ及其比值,采用RBC-C3b受体花环试验及RBC-IC花环试验检测红细胞免疫功能。结果:模型组大鼠血清IFN-γ/IL-4比值明显增高(P<0.01),RBC-C3b受体花环率明显低下(P<0.01),RBC-IC花环率明显升高(P<0.01)。各给药组均能明显纠正上述异常指标,其中调肝脾方组改善作用最好,能恢复IFN-γ/IL-4比值以及RBC-IC花环率至正常(P>0.05)。结论:调肝脾方对佐剂大鼠细胞因子的调节作用最显著,清除免疫复合物的能力最强。

Objective： To explore adjusting liver and spleen prescription and tonifying liver and kidney prescription on erythrocyte immune and eytokines of adjuvant rats by observing the serum cytokines interleukin-4（IL-4）,interfer- on-γ（IFN-γ） and the ratio and erythrocyte immune function. Methods： According to body weight,Wistar rats were randomly divided into normal group,model group,adjusting liver and spleen prescription group,tonifying liver and kidney prescription group,torch-gen-chip group,fenbid group. The autoimmune arthritis rats were induced by proinflammatory adjuvant method. The serum IL-4,IFN-γ were detected by ELISA method,and erythroeyte immune function were tested by RBC-C3b rosette and its receptors. Results： The serum IFN-γ/IL-4 ratio of the model group was signifieandy inereased（P〈0.01）. RBC-C3b rosette receptor rate was significantly lower（P〈0.01）,RBC-IC rosette rate significantly increased（P〈0.01 ）. Each treating group could significantly improve these indexes,and the adjusting liver and spleen prescription group had the best effeetion to recover the ratio of IFN-γ/IL-4 to resume and to lower RBC-IC rosette rate. Conclusion： Adjusting liver and spleen prescription has signifieent effects on regulating eytokines and clearing away the harmful immune complexes of adjuvant rats.
