摘要
目的:培养大鼠血管平滑肌细胞。方法:手术显微镜下分离主动脉和肺动脉,再用翻转干涸法进行操作。结果:显微镜下分离组织,可基本除净动脉纤维脂肪层和外膜,再用刀片轻刮内膜,可保证所贴组织块为动脉中膜。传至第三代时,用台盼蓝检查细胞传代成活率95%,光镜、电镜和免疫组化鉴定培养细胞,培养细胞纯度96%。结论:贴块片简便易行、经济,又可以防止膜损伤,可为血管病理变化研究提供适宜的细胞。
Objective: To culture rat vascular smooth muscle cell (VSMC). Methods: Pulmonary artery were separated carefully and cut into fine tissue pieces. These pieces used for culture of VSMC by method of adhesion to wall. Results: Cell cultured were typical form of VSMC under light microscope, electron microscope and immunohistochemical test. The cell viability is 95 % in subculture by trypan blue. The cell purity is 96% by immuohistochemical method. Conclusion: This method is simple and convenient. It provides ideal materials for vascular physiopathologic studies.
