摘要
改良 MTT法克服了常规 MTT法存在的缺陷。试验表明 ,用 1 0 % SDSp H4.5~ 4.7;每孔溶液总量控制在 30 0μl以内 ;每孔 1~ 2× 1 0 4TF- 1细胞是比较适宜的。本文探讨了生物学测定中误差的来源 ,采用增加不同时间的测定次数可有效降低测定误差。本法具有溶解彻底、颜色稳定、操作方便、结果准确、重现性好的优点 。
The defects in traditional MTT were overcome by improved MTT method. This results showed that 10% SDS with pH4.5~4.7 dissolved the formazan completely, that it is necessary and essential to control the total liquid volume within 300μl in every micro hole and that 1~2×10 4 TF 1 cell a well is suitable. The source of error in cytokins bioassay is also discussed in this paper. The advantages of improved MTT such as dissolveing completely, color stability, simplity of operation etc. so it is applicable for cytokine bioassay.
