转染B7-1基因至鼻咽癌细胞株及其诱导的细胞免疫应答 被引量:3

摘要 通过包装细胞把逆转录病毒载体pLXSN/B7-1导入鼻咽癌细胞株CNE1,利用RT-PCR及FACs等技术证实B7-1的表达情况,同时检测了CNE1细胞表面MHCI抗原。在此基础之上,观察了表达B7-1的CNE1细胞(CNE1/B7-1)对健康个体PBL(外周血淋巴细胞)的增殖及细胞因子分泌的影响,结果表明:CNE1/B7-1可以促进PBL的增殖及细胞因子的分泌,且增高水平的细胞因子主要是IL-2和IFN-γ。提示:CNE1/Β7-1诱导的免疫应答以细胞免疫为主。 The nasopharyngeal carcinoma cell strain CNE1 was transfected with retrovirus vector pLXSN containing the B71 cDNA sequence.The expression of B71 was evaluated by the technique of RTPCR and FACs,and the expression of MHC I was also analyzed.Furthermore we observed that the B71 expressing CNE1 not only could increase the proliferation of PBL from health donors,but also enhance secretion of some cytokines,the enhanced cytokines were mainly IL2 and IFNγ which typically secreted by the Th1 cells.This results indicate that transfected CNE1 cells mainly induce cellular immunity.
