摘要
通过对27个已确定HLA-DRB1基因型别的LCL进行TNF生物活性的检测,结果发现:TNF的表达水平与特定的HLA-DRB1等位基因型别关联,携带HLA-DRB1*070X的个体,其LCL经LPS刺激后,TNF生物活性水平比不携带HLA-DRB1*070X的个体低(P<0.01),且以HLA-DRB1*070X纯合子细胞表达量最低,杂合子次之。提示:HLA-DRB1遗传多态性参与调控LCL的TNF表达水平,影响TNF的产生可能是HLA复合体发挥免疫调节的重要因素之一。
The cytotoxicity of 27 HLAdefined normal LCL supernatants against TNFsensitive cell line L929 was tested.The results showed that culture supernatants of LPSstimulated LCLs of HLADRB1070X genotype individuals had a significantly lower TNF bioactivity than those from nonHLADRB1070X genotype individuals (P<0.01)esp.a lowest in DRB1070X homozygous cell lines.These findings suggest that TNF is one of the important factors participating in regulation of the immune response controlled by MHC,and variation of TNF production among individuals may be genetic trait associated with HLA polymorphism.
