老年男性动脉硬化及骨质疏松症与AHSG基因多态性的关系 被引量:1

The relationship of atherosclerosis and osteoporosis and polymorphisms of Alpha2-HS glycoprotein in elderly male

摘要 目的探讨老年男性α2-HS糖蛋白(AHSG)基因多态性与动脉硬化、骨质疏松、血脂以及血清骨相关生化指标的关系。方法用ELISA法测定208名老年男性患者的骨特异性碱性磷酸酶(BAP)、I型胶原羧基末端肽、I型胶原氨基末端肽和护骨素及瘦素的浓度;对患者基因组的DNA样品作限制性内切酶SacI的PCR-RFLP检测以确定其基因型,全自动生化仪酶法测定血清TC、TG、HDL-C、LDL—C,电极法测定血钙,DXA测定腰椎正位、仰卧侧位腰椎和股骨颈的骨密度(BMD)。GG基因型患者做骨组织病理活检,彩色超声诊断仪测定颈动脉内中膜厚度(IMT)。结果(1)老年男性患者AHSG基因CC型,CG型和GG型分布频率分别为60.2%,26.2%和13.6%。不同基因型血脂、血钙和骨生化指标有明显差异;(2)协方差分析显示老年男性不同AHSG基因型在正侧位腰椎、股骨颈的BMD和IMT差异有显著统计学意义;(3)GG型老年男性患者骨组织病理切片和彩超验证了AHSG多态性基因变异与动脉硬化、骨质疏松发生发展的相关性。结论AHSG基因多态性变异与老年男性血脂、血钙、血清骨生化指标差异,以及动脉硬化和骨质疏松的发病密切相关。 Objective To explore the relationship of Alpha2-HS glycoprotein (AHSG) gene polymorphisms and atherosclerosis and osteoporosis, and changes in blood lipids and serum bone relative biochemical markers in elderly male. Methods Blood samples of 208 elderly male inpatients were collected. Serum bone alkaline phosphatase (BAP), crosslinked N-telopeptide of collagen type I ( NTX), cross-linked C-telopeptide of collagen type I ( CTX), osteoprotegrin (OPG) and leptin were determined by ELISA. Serum TC, TG, HDL-C, LDL-C and calcium content were detected. AHSG gene polymorphisms were detected by polymerase chain reaction fragment length polymorphisms(PCR-RFLP) of restriction enzyme Sac I . BMD of the anteroposterior spine (AP), supine lateral spine (Lat), and femoral neck (FN) were measured by DXA( Norland XR-36). Morphological changes in the aorta and bone of GG genotypic patient were detected by pathological histology. IMT was measured by Color Doppler Ultrasound equipment (SEQUOIAS12). Results ①The genotype frequencies of CC, CG and GG were 60. 2%, 26. 2% and 13.6% in all elderly males respectively. There were significant differences in blood lipids, calcium and serum bone relative biochemical markers to various AHSG genotype.②There were significant differences in the BMD of AP, Lat, FN and IMT and the serum biochemical markers among CC, CG and GG genotypes. ③Bone tissue pathology histology in GG genotype of elderly males verifies AHSG polymorphism genetic mutation and atherosclerosis, osteoporosis development of the relationship. Conclusion Our study shows that the closely related among AHSG polymorphism variation and blood lipids, blood calcium, serum biochemical indicators of bone differences, and the incidence of artheriosclerosis and osteoporosis in elderly males.
