摘要
[目的]探讨邻苯二甲酸(2-乙基已基)(Diethylhexyl phthalate,DEHP)对小鼠雄性生殖系统的毒性作用机制。[方法]昆明种雄性小鼠(20±2)g,40只,随机分为4组,每组10只。经食饵自然给食染毒,连续4周后处死。观察小鼠体重及睾丸变化,测定不同剂量DEHP染毒小鼠的血液、睾丸中谷胱甘肽过氧化物酶(GPX)活性、一氧化氮(NO)和过氧化氢(H2O2)含量。[结果]DEHP染毒后,随着染毒剂量增加,小鼠体重逐渐减少(P<0.01),高剂量组睾丸脏器系数显著下降(P<0.01)。光镜下可见高剂量组睾丸组织有明显损伤。与对照组相比,DEHP中、高剂量暴露组小鼠血液及睾丸中GPX活性降低(P<0.01);H2O2含量增加(P<0.05,P<0.01);睾丸中NO含量降低,尤其是高剂量组与对照组差别明显(P<0.05,P<0.01)。[结论]DEHP对小鼠雄性生殖系统具有明显的毒性作用,其作用机制可能与氧化应激反应和NO含量降低有关。
[ Objective] To detect the mechanism of male preconception injuries induced by diethylhexyl phthalate(DEHP)in mice. [ Methods] Forty of Kumming mice (20 ± 2 )g were divided into 4 groups randomly, every group including 10 mice. The animals were exposed to DEHP by food for 4 weeks. At the end of the fourth week, they were terminated. Then calculated the weight and coeffcient of testis/body. GPX,NO,H2O2 of blood and testis were examined. [ Results] With the increase of DEHP ,the weight of mice and the coeffcient of testis/body with the high dose group decreased significantly(P〈 0.01 ). There were evident testes tissue damage in high dose group with examination by light microscope. Comparing with the control group,GPX activity in the blood and testes decreased dramatically (P 〈 0.01 ), while H2O2 content increased significantly (P 〈0. 05 ,P 〈0.01 ); in modprote and high dose qroups NO content decreased dramatically in testes,especially in high dose of DEHP( P 〈 0.05 ). [ Conclusion ] DEHP has significant male preconception injuries effect to mice. Thepossible mechanisms are oxidative stress injuries and NO content decrease.
