摘要
[目的]探讨错配修复蛋白hMSH2与白血病耐药发生的关系。[方法]MTT检测K562/ADM细胞耐药倍数,PT-PCR检测K562与K562/ADM的hMSH2基因表达,Western-blot检测二者的hMSH2蛋白表达,免疫细胞化学方法检测正常血有核细胞的hMSH2蛋白表达。[结果]K562/ADM耐药倍数为6.7倍,正常血有核细胞未见hM-SH2蛋白表达,Western-blot显示K562/ADM的hMSH2蛋白表达强于K562。[结论]白血病细胞K562存在hM-SH2基因及其产物的表达;耐药细胞的hMSH2蛋白增多可能与持续阿霉素作用有关。
[ Objective] To detect the expression of mismatch repair protein hMSH2 in the leukemia cell line K562 and its adriamycin -resistant cell line K562/ADM, and analyze the relation of drug resistance of leukemia to the hMSH2 protein expression [ Methods] MTT method was used to detect a multiple drug resistance of K562/ADM, RT - PCR to assay hMSH2 gene expression in K562/ADM and K562 cells, and Western - blot examin hMSH2 protein expression of" K562 and K562/ADM ceils. [ Resultsl K562/ADM resistance multiples was 6.7 times. Both of the K562 and K562/ADM cells have hMSH2 gene and protein expression, hMSH2 protein expression in K562/ADM was stronger than that of K562. [ Conclusion] It was not found that the peripheral blood nuclearcyte express hMSH2 protein, While the leukemia cell line express hMSH2 gene and its protein. The hMSH2 protein maybe participate in adriamycin resistance of leukemia.
