摘要
目的:观察仙草体外抗乙型肝炎病毒(HBV)的活性。方法:以HepG2.2.15细胞株为模型,用微粒酶免疫测定技术(MEIA)检测细胞培养上清液中HBsAg、HBeAg含量,用PCR荧光定量技术检测细胞上培养上清液中HBV DNA的变化,以MTT比色法观察药物的细胞毒性。结果:仙草对HepG 2.2.15细胞的TC50为37.45 mg/mL,对抑制HepG2.2.15细胞分泌HBsAg和HBeAg的治疗指数分别为14.71和39.42。对HBV-DNA仅有微弱抑制作用。结论:仙草在体外细胞培养中具有直接抗HBV活性。
Objective: To evaluate the inhibitory effects of Xiancao(hsian-tsao,Mesona proeumbensHemsl) against hepatitis B virus (HBV)in vitro. Methods: HepG2.2.15 cells, as model cells, were exposed to different concentrations of extracts of Xiancao(hsian-tsao,Mesona procumbensHemsl) and IF α 1b, respectively. MEIA was used to assess HBsAg and HBeAg; fluorescent quantitative PCR was applied to quantify extraeellular HBV-DNA, MTT chromatogramphy was applied to observe toxicity . Results: the TC50 of Xiancao(hsian- tsao,Mesona procumbensHemsl) is 37.45 mg/mL, TI against HBsAg, HBeAg are 14.71,39.42 respectively, and also Xiancao inhibits HBV replicationin vitro in to some extent. Conclusion: Xiancao have the anti-HBV activity in vitro.
