五指山猪H-FABP基因与生长性状的关联分析 被引量:3

摘要 以五指山猪为试验材料,采用PCR-SSCP和测序技术相结合,对五指山猪心脏脂肪酸结合蛋白(H-FABP)基因进行多态检测,并分析其与生长性状(体质量、体高、体长、胸围)的相关性。在五指山猪H-FABP基因Exon2中发现了3种基因型,分别命名为AA、AB、BB,对其中的纯合子AA、BB进行测序,发现Exon2的第30、120位点处发生了突变,都是由碱基C→T的转换造成的,并且导致脯氨酸突变为亮氨酸,苏氨酸突变为异亮氨酸。AA、AB、BB三种基因型频率分别为15.2%、66.7%、18.1%,A、B等位基因频率分别为48.6%和51.4%。不同基因型与生长性状的方差分析表明,8月龄体质量、体长和10月龄胸围AB与AA、BB基因型差异显著(P<0.05),其余分析无显著差异(P>0.05)。 Applying the PCR-SSCP combined with sequencing techniques, the H-FABP gene in WZS pig was analyzed in the trial based on WZS pigs and the correlation with growth traits (body weight, body height, body length and chest circumference) was analyzed as well. The results indicated that 3 genotypes were discovered in Exon2 and respectively defined as AA,AB and BB. Through the sequencing analysis of the two homozygous genes-AA and BB, the mutation of the 30th and 120th polymorphic sites of the Exon2 of porcine H-FABP gene was discovered. Both mutation was resulted from C →T transition and induced that proline was transferred to be leucine and threonine to be isoleucine. The frequencies of AA, AB and BB were respectively 15.2%, 66.7% and 18.1% ; A and B allele frequencies were 48.6 % and 51.4%. ANOVA of different genotypes of H-FABP gene sites and 4 traits showed that there were significant differences in 8-month-old of body weight, length (P〈0.05) and 10-month-old chest circumference (P〈0.05) between AB,AA and BB genotypes, but there was no significant difference in other-month-old growth traits among other genotypes.
