摘要
目的:建立补肾排毒颗粒中黄芪甲苷的HPLC-ELSD测定方法。方法:色谱柱:ZORBAX SB-C_(18)柱(250mm×4.6mm,5μm);流动相:乙腈-水(35:65);柱温:30℃;流速:1.0 ml·min^(-1);ELSD检测器漂移管温度:105℃,氮气流速:2.7ml·min^(-1)。结果:黄芪甲苷在2～30μg范围内呈良好线性关系(r=0.999 6),平均回收率为97.5%,RSD为1.2%(n=6)。结论:方法简便、结果准确,可用于该制剂的质量控制。
Objective： To establish a HPLC-ELSD method for determination of astragaloside Ⅳ in Bushenpaidu granules. Method： The Zorbaxs-C18 column （ 250mm × 4. 6mm,5 μm） was used, the mobile phase was aeetonitrile-water （ 35：65 ） , the column temperature was 30℃, the flow rate was 1.0 ml · min ^-1, ELSD as the detector, the temperature of the drift tube was 105℃, the flow rate of N2 was 2.7L · min ^-1. Result： The calibration curve was linear within the range of 2-30μg for astragaloside Ⅳ （ r = 0. 999 6 ）, the average recovery was 97.5 %, RSD = 1.2 % . Conclusion ： The method is simple, sensitive and accurate, it can be used for quality control of Bushenpaidu granules.
