下调表达VEGFR-2受体对神经胶质瘤U87细胞生物学行为的影响

Effects of VEGFR-2 siRNA on biological characteristics of VEGFR-2 receptor-positive U87 cells

摘要 目的观察沉默血管内皮生长因子受体2(VEGFR-2)基因后对表达VEGFR-2受体的U87细胞生物学行为的影响。方法合成VEGFR-2小干扰RNA(si RNA),转染人U87细胞,通过适时定量PCR检测筛选出最有效VEGFR-2si RNA后,进行Western blot验证。转染的U87细胞,通过流式细胞术检测评价细胞凋亡,Transwell实验评价侵袭能力和MTT检测细胞增殖能力。结果成功筛选出有效VEGFR-2siRNA,体外研究,将其稳定转染人U87细胞。与空白对照组、阴性对照组相比,U87细胞表现出增殖能力和侵袭能力均下降,但未见明显细胞凋亡改变。结论抑制U87细胞自身表达的VEGFR-2可有效地抑制其生长,降低侵袭能力。 Objective To study the effects of vascular endothelial growth factor receptor (VEGFR)-2 small interfering RNA (siRNA) on biological characteristics of VEGFR-2 receptorpositive U87 cells. Methods VEGFR-2 siRNA was synthesized and transfected into U87 cells. The most effective VEGFR-2 siRNA was selected by real-time quantitative PCR and verified by Western blot. The apoptosis, invasion and proliferation of transfected U87 cells were evaluated with flow cytometry, transwell and MTT. Results The most effective VEGFR 2 siRNA was successfully screened and transfected into U87 cells in vitro. Compared with the negative control group and control group, VEGFR-2 siRNA led to suppression of U87 cells growth, reduction of invasion, but not increase of apoptosis. Conclusion Inhibiting the self-expression VEGFR-2 of U87 cells could effectively inhibit the growth of U87 cells and reduce the invasion.
