有丝分裂阻滞缺陷蛋白2(MAD2)在哺乳动物卵母细胞减数分裂成熟中的作用 被引量:1

摘要 有丝分裂阻滞缺陷蛋白2(mitotic arrest deficient2,MAD2)为一种纺锤体检验点蛋白,是监控卵子减数分裂纺锤体行为和染色体分离的最重要的关卡蛋白之一。人MAD2编码一段比较保守的Mr=24000蛋白片段,它的单倍体和寡聚体状态、非磷酸和磷酸化状态与它的活性密切相关。MAD2的定位在不同的种属、不同的生殖细胞、不同的细胞分裂状态间都有差异。在卵母细胞中,MAD2细胞定位(胞质定位和动粒定位)随减数分裂细胞周期的进程和/或微管与动粒之间的联系改变而发生相应的变化。在哺乳动物卵母细胞减数分裂成熟过程中,MAD2的主要作用是控制后期的起始和染色体的分离;另外,它与卵母细胞的凋亡有关,并可能参与卵母细胞减数分裂的恢复。目前已有的2种关于纺锤体检验点的作用机制模式,但都存在缺陷。
