摘要
目的:探讨地塞米松(DEX)对顺铂(DDP)诱导卵巢癌细胞株凋亡的影响。方法:应用MTT比色法检测细胞活性,利用免疫组织化学法检测凋亡相关基因bcl—xl和caspase3的表达。结果:0.001~1μM浓度范围内的地塞米松对顺铂诱导人卵巢癌细胞株HO一8910的凋亡有抑制作用(P〈0.05),提高DDP浓度能拈抗该效应;并能上调抗凋亡蛋白bcl—xl的表达,降低caspase3的表达活性(P〈0.05)。结论:顺铂治疗卵巢癌前用地塞米松进行预处理后可拮抗顺铂的抗肿瘤作用,bcl—xl抗凋亡通路参与该作用。
Objective: To explore the effect of dexamethasone on cisplatin (DDP) - induced apoptosis of ovarian cancer cells. Methods: Cellular activities were detected by MTT colorimetry , and the expression levels of bcl - xl and caspase3 protein were analyzed by immunocytochemistry. Results: 0. 001 - 1μM dexamethasone inhibited the apoptosis of HO - 8910 cells induced by DDP (P 〈 0.05 ), and this effect weaken with the increasing of DDP ; additionally, it up - regulated the expression of bcl - xl and down - regulated the activity of caspase- 3 protein (P 〈 0. 05 ) . Conclusion: Pretreatment of human ovarian cell lines HO -8910 with dexamethasone can inhibit the anti - tumor effect of DDP, and the anti - apoptosis pathway of bcl - xl is involved.
