摘要
目的研究乙胺丁醇在乙胺丁醇-磷酸钙骨水泥复合体(Ethambutol-calcium phosphate cement-compound E-CPC-C)中缓慢释放的效果。方法以白兔为实验对象,分为臀肌组和股骨组,将E-CPC-C植入兔臀大肌中和股骨髓腔中,以质谱仪测定其植入后一定时间的血药浓度及局部组织的药物浓度。结果臀肌组血药浓度在E-CPC-C植入后第2~7d达0.5μg/ml以上,股骨组血药浓度仅在植入后第4d达0.56μg/ml,植入后第4周E-CPC-C周围组织药物浓度臀肌组达1.6μg/g,股骨组达2.2μg/g。结论E-CPC-C能在体内持续、缓慢释放乙胺丁醇,对治疗骨结核可能是一有效植入式缓释剂。
Objective To investigate whether ethambutol was slowly released by ethambutol-calcium phosphate cement-compound(E-CPC-C) in vivo. Methods Rabbits were divided into the gluteus grloup and the femurs group.E-CPC-C were implanted into the gluteus maximus or the distal metaphysis of femurs of rabbits,Ethambutol concentration in blood and in tissue around E-CPC-C at different time after implantion was analyzed by mass spectrograph.Results Ethambutol concentration in blood of the gluteus grloup was over 0.5μg/ml at the 2th-7th day,and the femurs group's was 0.56μg/ml only at the 4th day, ethambutol concentration in local muscle tissue was 1.6μg/g at the 4th week, 2.2μg/g in local bone tissue. Conclusion E-CPC-C can slowly release ethambutol in vivo. It may be an effective implanting sustained biomaterial to treat bone tuberculosis.
