摘要
目的:探讨大鼠肝脏纤维化损伤后血浆白蛋白mRNA水平、丙氨酸转氨酶、羟脯氨酸的活性变化用于评估早期肝损伤的可能性。方法:建立大鼠肝纤维化模型,在不同时间分离血清,采用实时定量多聚酶链反应技术测量血浆中白蛋白的mRNA水平;生化检测血浆丙氨酸转氨酶(ALT)、羟脯氨酸(Hyp)活性。结果:血浆中白蛋白mRNA水平在肝纤维化早期显著增加,丙氨酸转氨酶、羟脯氨酸在肝纤维化早期变化不明显。结论:在大鼠慢性肝损伤早期检测血浆中白蛋白mRNA比丙氨酸转氨酶、羟脯氨酸活性更具敏感性。
Objective: To investigate serum albumin (AST) and Hydroxyproline(Hyp) in injury of rat liver fibrosis. mRNA level, activity of alanine aminotransferase Methods: The rat liver fibrosis model was created by injection Bovine Serum Albumin(BSA), separated from the serum at different times, using real-time quantitative Polymerase Chain Reaction technique measured the plasma level of albumin mRNA, then detection activity of plasma ALT and Hyp by chemical and biological technique. Results: The plasma level of mRNA was detected a significant increase in the early stage of liver fibrosis, ALT and Hyp were not significantly changed in the early stage of liver fibrosis. Conclusion : Detection of serum Albumin mRNA has more sensitive activity than plasma Alanine Aminotransferase and Hydroxyproline in the early of chronic liver damage in rats.
