Selection of the optimal condition for determination of rutin concentration by spectrophotometry

摘要 利用总黄酮类化合物与铝离子生成有色络合物,其吸收强度与络合物浓度成正比来测定芦丁,从试验中选择对芦丁测定的最佳λ射光波长、pH值、显色时间、显色剂用量,并通过这4个因素的相互作用,利用正交试验得到最佳的分析条件为A1C3D2B1,即λmax=480nm,显色时间5m in^15min,显色剂用量为5mL,pH12为最佳组合条件。 Flavonoids and Al^3+ can form colored chelate compound, and its absorbency is proportional to the concentration of chelate compound. This was applied to determine the concentration of rutin by spectrophotometry. Optimal maximum wavelength, pH, developing time and dosage of developer were obtained through the tests. Furthermore, the orthogonal test was used to get the best combination of these four factors. Results is indicated : the best condition is A1 B1 C3 D2, which meant λmax =480nm developing time 5min - 15min, dosage of developer 5ml, pH 12.
