半乳糖特异性植物血凝素富集胎儿有核红细胞 被引量:1

摘要 目的建立半乳糖特异性植物血凝素分离和富集胎儿有核红细胞的方法和反应装置,为胎儿有核红细胞用于产前诊断提供基础。方法选用38~40孕周正常分娩胎儿脐血,经半乳糖特异性植物血凝素反应体系分离和富集胎儿有核红细胞,半乳糖浓度为50、100、100、150、200μg/mL、大豆凝集素浓度为10、20、30、40、50μg/mL。特异性荧光抗体染色后用激光共聚焦显微镜观察并计数分离和富集到的胎儿有核红细胞。结果半乳糖特异性植物血凝素反应体系能够有效分离和富集胎儿有核红细胞,当包被玻片的半乳糖浓度为100μg/mL、大豆凝集素浓度为20μg/mL时,与有核细胞的比率最高为(6.54±0.56)%。结论半乳糖特异性植物凝素可用于选择性地分离和富集胎儿有核红细胞。 Objectives To construct the reaction system to isolate and enrich fetal nucleated red blood cells(FNRBCs) by using galactose specific lectin. Methods Using the density gradient centrifugation to isolate the needed cells from the cord blood, then grading-up the condition of the reaction system by using laser scanning confocal microscope. Results The glass slid was coated with certain concentration of GalNAc was 50,100,100,150,200μg/mL, and integrate the detected cells with SBA was 20μg/mL, the rate of FNRBCs to nucleus cells was the highest(6.54±0.56) %. Conclusion Galactose-specific lectin can be used to effectively isolate and enrigh FNRBCs.
